Archiving and rederivation of mouse mutants

  • cryopreservation of germ cells and embryos
  • steril embryotransfer into isolators, IVC-cages or SPF-units

The ÖZBT will provide the generation and the breeding of mouse mutants exclusive under SPF-conditions. Therefore, the import of mice with undefined or unknown hygienic quality into our facilities is performed only via embryo transfer. Additionally, we can accept germ cells or embryos instead of life animals from other laboratories. Archiving of mouse mutants is possible by cryopreservation of germ cells and embryos. Exceptionally, we will offer the preservation of embryos. The main focus concerning archiving is on the cryopreservation of sperm cells. The ability of mouse sperm cells to fertilize oozytes in vitro is affected by several technical factors as well as by the genetic background of the mutant. Therefore, we intend to analyse and to optimize the method for each genetic background and in order to achieve the best results for in vitro fertilization (IVF).  Additionally, we plan to establish intracytoplasmatic sperm injection (ICSI). All methods of assisted reproduction are also usable in case of problems in reproduction of a mutant.
